Characterization of two bacterial mutants with temperature-sensitive synthesis of DNA.

چکیده
DESPITE considerable interest in recent years, the replication of bacterial DNA and the mechanism of its regulation are still poorly understood, as can be seen in recent reviews of the subject (LARK 1966; MAALBE and KJELDGAARD 1966). One approach to the study of essential cellular syntheses is through the study of temperature-sensitive mutants (APIRION 1966; EPSTEIN et al., 1963; NEIDHARDT 1966). Already several papers have appeared describing bacterial mutants impaired in synthesis of DNA at higher temperature ( BONHOEFFER 1966; KOHIYAMA et al. 1966; MENDELSON and GROSS 1967). The present report describes a procedure for the isolation of bacterial mutants with tcmperaturesensitive synthesis of DNA and the characterization of two mutants representative of the types which were most frequently encountered. The mutants studied resemble the two classes found previously (KOHIYAMA et al. 1966); but from the experiments presented here, it is evident that at least one of them represents a new kind of block in DNA synthesis. A preliminary report of these mutants has been presented ( FANGMAN 1966).

Selection and preliminary characterization of temperature-sensitive mutants of type 5 adenovirus.
Eight temperature-sensitive (ts) mutants that replicate normally at 32 C but poorly, if at all, at 39.5 C have been isolated from mutagenized stocks of a wild-type strain of type 5 adenovirus. Three mutagens were employed: nitrous acid, hydroxylamine, and nitrosoguanidine. Ts mutants were isolated from mutagenized viral stocks with frequencies between 0.01 and 0.1%. All eight mutants had revers...
متن کاملIsolation of temperature-sensitive cell cycle mutants from mouse FM3A cells. Characterization of mutants with special reference to DNA replication.
A large number of mutants that are temperature sensitive (ts) for growth have been isolated from mouse mammary carcinoma FM3A cells by an improved selection method consisting of cell synchronization and short exposures to restrictive temperature. The improved method increased the efficiency of isolating DNA ts mutants, which showed a rapid decrease in DNA-synthesizing ability after temperature ...
متن کاملNovel Pt(II) Complex and Its Pd(II) Aanalogue. Synthesis, Characterization, Cytotoxicity and DNA-interaction
The ability of small molecules to perturb the natural structure and dynamics of nucleic acids is intriguing and has potential applications in cancer therapeutics. This work reports the synthesis, characterization, cytotoxicity and DNA-binding studies of two cytotoxic and intercalative [M(bpy)(pyrr-dtc)]NO3 complexes (where M = Pt(II) and Pd(II), bpy = 2,2´-bipyridine and pyrr-dtc = p...
متن کاملLarge-scale automated isolation of Escherichia coli mutants with thermosensitive DNA replication.
We have screened about 1.4 million colonies of Escherichia coli K-12 for their ability to grow on nutrient agar at 30 degrees and 41 degrees. Among the 2266 temperature-sensitive mutants found, 110 were defective in DNA synthesis but not in protein synthesis at 41 degrees. Three of these dna mutations mapped at two previously undescribed loci on the E. coli genetic map and may represent new gen...
متن کاملTemperature-sensitive mutants of a Chinese hamster cell line. I. Selection of clones with defective macromolecular biosynthesis.
Temperature-sensitive clones have been selected from a mutagenized culture of Chinese hamster lung cells by a procedure involving bromodeoxyuridine (BrdU) incorporation and irradiation with black light. The selection procedure used in these studies was adapted from methods developed by others to yield mutants that cease DNA replication within a short time after they are transferred to nonpermis...
